Solubility of DL-Methionine
Water solubility: DL-methionine isslightly soluble in water, which is a typical characteristic of non-polar amino acids. At 25 °C, its solubility in pure water is approximately3.3 g/100 mL. The solubility increases with the rise of temperature—at 80 °C, the solubility can reach about 10 g/100 mL. The aqueous solution of DL-methionine is neutral, with a pH value of 5.6–6.2, which is close to the isoelectric point (pI=5.74) of methionine; at the isoelectric point, DL-methionine has the lowest solubility due to the minimal degree of molecular ionization.
Solubility in organic solvents: DL-methionine shows different solubility in organic solvents due to the presence of polar functional groups (-NH2,-COOH) and non-polar side chains (-CH2CH2SCH3) in its molecule. It isslightly soluble in methanol and ethanol(solubility < 1 g/100 mL at 25 °C), andalmost insoluble in non-polar solventssuch as ether, chloroform, benzene, and petroleum ether. This solubility feature enables the separation of DL-methionine from organic solvent-soluble impurities through solvent extraction.
Solubility in acid-base solutions: DL-methionine is readily soluble in dilute acid and alkaline solutions. In 0.1 mol/L hydrochloric acid or sodium hydroxide solution, its solubility is significantly increased to more than 20 g/100 mL, because the amino group and carboxyl group in the molecule are ionized in acid-base environments, enhancing the hydrophilicity of the molecule.
