In the field of peptide synthesis, BOC-L-GLU(ME)-OME is the standard building block for peptides containing glutamic acid residues, mainly used in solid-phase and liquid-phase synthesis. The Boc protecting group can be easily removed under acidic conditions, and the methyl ester side chain remains stable under condensation conditions, allowing for precise control of the peptide chain extension sequence and avoiding side-chain carboxyl side reactions. It is widely used in the preparation of antimicrobial peptides, neuroregulatory peptides, enzyme inhibitors, and targeting peptides.
In the development of peptide drugs, BOC-L-GLU(ME)-OME can effectively improve coupling efficiency, reduce racemization rate, and ensure the uniformity of product configuration. When using it, the strength of deprotection must be strictly controlled to avoid peptide chain breakage or side-chain methyl ester hydrolysis due to excessive strong acid treatment. At the same time, the condensation reaction should be carried out in an anhydrous condition to prevent the failure of raw material hydrolysis.
